THE EXPRESSION OF TLR4, IFN-γ, TGF-β AND TNF-αLL LINE OF HUMAN SMALL CELL LUNG CARCINOMA NCI-H69 AND IN CISPLATIN-RESISTANT SUBLINE NCI-H69/CPR

Aim: to investigate the effect of teichoic acid Staphylococcus aureus for expression pro-inflammatory cytokines and TLR4 in a human small cell lung carcinoma line NCI-H69, cisplatin resistant subline NCI-H69/CPR. Methods. Incubation cells with (1 ng/m) conducted 2 days. Expression level TLR4, TGF-β, INF-γ, TNF-α was evaluated by real time PCR on 7500 Real-Time System, using specific primers fluorochrome SYBR Green. The reverse transcription reaction performed High-Capacity cDNA Reverse Transcription Kit carried out under conditions: 25 °C - 10 min, 37 ‒ 120 min 85 5 min. Results. In culture NCI-H69 addition increased 1.3 times, IFN-γ – 1,1 times. TGF-β decreased 2.5 4.9 times respectively. NCI-H69/CPR inhibited all studied parameters. 4.2 1.4 depressed 1.6 1.2 presented data indicate that bacterial origin provided modulating inflammatory cancer culture, sensitive cisplatin. Conclusions. Teichoic as ligand modulates anti-inflammatory suppresses investigated cisplatin-resistant NCI-H69.
